Understanding the ENERGY-3 Pivotal Clinical Trial for ENPP1 Deficiency

ENERGY-3 Pivotal Clinical Trial Webinar

Brighter Hope Foundation is committed to sharing timely, accurate information about research and clinical trials that may impact individuals and families affected by rare genetic calcification disorders. The ENERGY-3 Pivotal Clinical Trial Webinar provides an in-depth overview of an important late-stage clinical study evaluating a potential treatment for individuals living with ENPP1 Deficiency and related conditions.

What Is the ENERGY-3 Trial?

The ENERGY-3 trial is a pivotal clinical study designed to evaluate the safety and effectiveness of an investigational therapy targeting the underlying cause of ENPP1 Deficiency. ENPP1 Deficiency is a rare genetic disorder caused by mutations in the ENPP1 gene, which plays a critical role in regulating pyrophosphate (PPi), a molecule that helps prevent abnormal calcification in the body.

When ENPP1 function is impaired, pyrophosphate levels decrease. This can lead to abnormal calcium buildup in arteries and soft tissues. In infants, this may present as Generalized Arterial Calcification of Infancy (GACI), a life-threatening condition characterized by calcification and narrowing of large and medium-sized arteries. Over time, individuals with ENPP1 Deficiency may also develop complications such as Autosomal Recessive Hypophosphatemic Rickets Type 2 (ARHR2), which affects bone health and phosphate balance.

The ENERGY-3 trial focuses on addressing the root biochemical pathway involved in these conditions by aiming to restore the balance of pyrophosphate in the body.

Why This Webinar Matters

This webinar explains:

  • The scientific rationale behind the investigational therapy
  • How the treatment is designed to target the underlying mechanism of disease
  • The structure and goals of a pivotal Phase 3 clinical trial
  • Eligibility considerations and what participation may involve
  • How researchers measure safety and clinical benefit

For families navigating ENPP1 Deficiency, GACI, or ARHR2, understanding the science behind clinical trials can feel overwhelming. This webinar breaks down complex medical concepts into accessible information, empowering families to make informed decisions in partnership with their healthcare teams.

How This Relates to the GACI Community

ENPP1 Deficiency is one of the primary genetic causes of GACI, and research advancements in ENPP1-targeted therapies may directly impact outcomes for infants diagnosed with GACI as well as older children and adults living with long-term complications. Because Brighter Hope Foundation supports individuals affected by ENPP1 Deficiency, ARHR2, and other related calcification disorders, updates on pivotal research like ENERGY-3 are critical to our mission.

Clinical trials represent hope—but also important questions. Through educational webinars like this one, Brighter Hope Foundation continues to provide transparent, evidence-based information so families can better understand emerging treatment options and ongoing research efforts.

We encourage families, caregivers, and healthcare professionals to watch the full ENERGY-3 Pivotal Clinical Trial Webinar and discuss any questions about clinical trial participation with a qualified medical provider.
